12 weeks of NMN: NAD⁺ metabolism and arterial health

This 12-week, randomised, double-blind, placebo-controlled trial followed 36 healthy middle-aged adults (ages 40–59) taking 125 mg of NMN twice daily, 250 mg total per day.

NMN works through NAD⁺. Serum nicotinamide, a downstream marker of NAD⁺ metabolism, rose about 58% in the NMN group while falling in the placebo group, confirming that oral NMN actively feeds the NAD⁺ pathway in people.

Arterial stiffness. A measure of arterial flexibility (pulse-wave velocity) trended in a favourable direction with NMN, though it didn't reach statistical significance across the whole group.

Who benefited most. In participants with above-average BMI or blood glucose, NMN significantly improved arterial stiffness versus placebo, suggesting those under more metabolic strain may have the most to gain.

Safety. 250 mg/day was well tolerated over 12 weeks, with no adverse events and no concerning changes in blood chemistry or hormones.
